We Tested 30 Lion’s Mane Extracts — Here’s What We Found

Functional mushroom supplements are one of the fastest-growing categories in European health and wellness. But growth has outpaced transparency. Bold claims on labels. Impressive-sounding numbers on certificates of analysis. And very little independent verification of what’s actually inside the product.

We decided to test it. Not with supplier-provided paperwork — through an independent university research project analysing 30 lion’s mane extract samples sourced from suppliers across the global market.

The results were sobering. And they matter if you’re a brand, a formulator, or a buyer sourcing functional mushroom ingredients in Europe.

The Testing Methodology

All 30 samples were analysed as part of an independent research project conducted in partnership with Shannon ABC at the Technological University of the Shannon (TUS), and tested for:

  • Beta-glucan content — the primary bioactive polysaccharide in medicinal mushrooms
  • Hericenones C and D — the neuro-active compounds unique to lion’s mane, linked to nerve growth factor (NGF) stimulation
  • Triterpene profiles — secondary metabolites with anti-inflammatory and immunomodulatory properties

This wasn’t supplier-side quality control. This was independent university research with no commercial relationship to any of the suppliers.

Finding #1: Beta-Glucan Content Ranged from Below 5% to 82%

Across the 30 samples, beta-glucan readings ranged from below 5% to 82%.

That range alone should concern any buyer. But the high end is arguably more troubling than the low end.

Genuine lion’s mane fruiting body extracts do not typically exceed 30–40% beta-glucan content. Readings of 82% are not indicators of exceptional quality — they indicate manipulation or adulteration. The most common method is spiking with purified beta-glucan from non-mushroom sources, such as yeast or oats.

For B2B buyers, this means a certificate showing “60% beta-glucans” isn’t necessarily better than one showing 28%. It may simply be less honest.

Finding #2: Hericenones Not Detected in Any Meaningful Concentration

This was the most striking result.

Hericenone C and D — the neuro-active compounds that make lion’s mane unique — were not detected in any meaningful concentration in any of the 30 samples.

These are the compounds most commonly cited in lion’s mane marketing. “Supports cognitive function.” “Promotes nerve growth factor.” The science behind these claims rests largely on hericenones and erinacines. Yet the commercial extracts we tested showed no meaningful presence of these compounds.

This doesn’t mean the compounds don’t exist in lion’s mane. It means they’re either not surviving the extraction process, not present in sufficient concentrations in the cultivated material, or the growing and harvesting conditions aren’t optimising for these metabolites.

Finding #3: Only 2 of 30 Samples Showed Genuine Triterpene Identification

Triterpenes were detected across multiple samples. However, when subjected to high-confidence analytical identification, only two out of 30 samples showed genuine, verifiable triterpene profiles.

The rest showed interference, misidentification, or concentrations too low for confident attribution. In practical terms: most of these extracts could not substantiate triterpene claims under rigorous laboratory scrutiny.

The “Third-Party Tested” Problem

One of the most pervasive issues in functional mushrooms is the term “third-party tested.”

When a mushroom supplement brand says its product is third-party tested, the reality is often this: the raw material supplier provided a certificate of analysis confirming that the ingredient met specifications before it was shipped. That COA tests raw materials before formulation and packaging — not the finished product the consumer receives.

That is not independent testing. That is the person selling you the ingredient, marking their own homework.

True third-party testing means submitting your finished product to an accredited laboratory with no commercial interest in the result. It means testing for species-specific bioactive markers — not just total polysaccharides. And it means publishing the results.

Why Extraction Ratios Are Meaningless

Many mushroom extract suppliers market their products using ratios like “10:1” or “15:1,” implying higher concentration. But there is no standardised assay for extraction ratio. A 10:1 label tells you nothing about the actual bioactive content of the extract. It’s a production metric, not a quality metric.

What matters is what’s in the finished product: beta-glucan percentage (measured by validated methods), species-specific compounds (hericenones for lion’s mane, ganoderic acids for reishi, cordycepin for cordyceps), and contaminant testing (heavy metals, pesticides, microbial).

What Determines Extract Quality?

Our research, conducted in partnership with Emma Murphy, Senior Business Development Scientist at Shannon ABC, Technological University of the Shannon (TUS), identified several production factors that significantly affect extract quality:

Growing Medium

Mushrooms grown on natural substrates — hardwood logs for reishi, hardwood sawdust blocks for lion’s mane — produce measurably higher concentrations of bioactive compounds compared to grain-spawn cultivation in laboratory settings. Most commercial mushroom powders use grain spawn, which reduces cost but also reduces the bioactive profile.

Harvest Timing

Growth duration matters enormously. Reishi harvested at 5 months versus 11 months shows significant differences in triterpene and ganoderic acid concentration. Longer cultivation cycles are more expensive but produce substantially richer extracts.

Extraction Method

Many Chinese mushroom powders are steam-treated to meet export safety standards. Steam treatment does not constitute extraction, despite marketing claims. Genuine extraction involves hot water extraction, alcohol extraction (dual extraction), and increasingly, chromatographic purification that isolates specific bioactive compounds while removing impurities.

The Emerging Science: Golden Oysters and Ergothioneine

Beyond the current six-species lineup, we’re watching the science on golden oyster mushrooms (Pleurotus citrinopileatus) with particular interest. Golden oysters are exceptionally rich in ergothioneine — a unique amino acid antioxidant increasingly referred to as the “longevity vitamin.”

Early research suggests ergothioneine may play a role in cellular protection against oxidative stress and age-related decline. As analytical methods improve and clinical data accumulates, we expect golden oysters to become a significant addition to the functional mushroom category.

What This Means for B2B Buyers

If you’re sourcing functional mushroom extracts for your brand, the takeaway is straightforward:

  1. Don’t rely on supplier COAs alone. Test your finished products independently, in ISO 17025-accredited labs.
  2. Be sceptical of unusually high beta-glucan claims. If it sounds too good to be true, it probably is.
  3. Ask for species-specific compound testing. Total polysaccharides tell you almost nothing useful.
  4. Ignore extraction ratios. They’re not verifiable and not meaningful.
  5. Demand traceability. Know where the mushroom was grown, on what substrate, for how long, and how it was extracted.

The functional mushroom market in Europe is projected to grow at over 11% CAGR through 2031. Brands that invest in genuine quality infrastructure now will be the ones still standing when regulatory scrutiny inevitably increases.
